Genome‐wide identification of transcription factor binding sites with the ChIP‐seq method is an extremely important scientific endeavor − one that should ideally be performed for every transcription factor in as many cell types as possible. A major hurdle on the way to this goal is the necessity for a specific, ChIP‐grade antibody for each transcription factor of interest, which is often not available. Here, we describe CETCh‐seq, a recently published method utilizing genome engineering with the CRISPR/Cas9 system to circumvent the (...) need for a specific antibody. Using the CETCh‐seq method, targeted genomic editing results in an epitope‐tagged transcription factor, which is recognized by a well‐characterized, standard antibody, efficacious for ChIP‐seq. We have used CETCh‐seq in human cancer cell lines as well as mouse embryonic stem cells. We find that roughly 60% of transcription factors tagged using CETCh‐seq produce a high quality ChIP‐seq map, a significant improvement over traditional antibody‐based methods. (shrink)

Advertising and other forms of promotional activity have proliferated to such an extent that they may constitute a form of social pollution (Kitchen, 1994). The quantity and tone of communications to which consumers are exposed may have a subtle but pervasive effect on the social ecology of the developed world. Not only are Marketing Communications delivered in unprecedented quantities (Kitchen, 1994); but their tone is increasingly difficult to categorise in the Postmodern Marketing era (Brown, 1994). Notably, there has been very (...) little research conducted on this seeming "Leviathan" (Kitchen, 1994, after Hobbes, 1651) effect. Ethical concerns of professional marketing bodies such as the MRS and CIM are focussed on the conduct of professionals with regard to law and notions of moral decency in human exchange relationships (see MRS code of conduct, ASA and IBA guidelines). There is less emphasis on the ethical implications for society of the totality of Marketing Communication activities. This paper examines the distinctively Postmodern concept of the Communications Leviathan and discusses contemporary ethical issues, drawing perspectives where possible from Postmodern critical theory, Enlightenment philosophy, cognitive psychology and classical ethical works of Plato and Aristotle. Five appropriate roles of professional philosophers in environmental scholarship, policy and education are explored. Philosophers can and should function as social critics, as conceptual analysts, as “critical spectators” of science, as ethicists, and as educators. An example of the philosopher's role as social critic may be found in the analysis of the methods and presuppositions of public policy making. The philosopher as conceptual analyst can add clarity to environmental debates through analyses of such terms as “ecosystem”, “integrity”, “responsibiity” and “nature”. (...) “Meta-Science” is exemplified in an examination of the role of values in scientific investigation and in the scientist's contributions to public policy-making. The philosopher as environmental ethicist should seek a balanced and integrated view of both the naturalness of the human condition, and of the unique human quality of moral agency. Finally, all these contributions, and more, are combined in the philosopher's function as educator—first, of his student, and also of his profession, his academic colleagues, public policymakers, and the public at large. Can the use of psychedelic drugs induce lasting changes in metaphysical beliefs? While it is popularly believed that they can, this question has never been formally tested. Here we exploited a large sample derived from prospective online surveying to determine whether and how beliefs concerning the nature of reality, consciousness, and free‑will, change after psychedelic use. Results revealed significant shifts away from ‘physicalist’ or ‘materialist’ views, and towards panpsychism and fatalism, post use. With the exception of fatalism, these changes endured (...) for at least 6 months, and were positively correlated with the extent of past psychedelic‑use and improved mental‑health outcomes. Path modelling suggested that the belief‑shifts were moderated by impressionability at baseline and mediated by perceived emotional synchrony with others during the psychedelic experience. The observed belief‑shifts post‑psychedelic‑use were consolidated by data from an independent controlled clinical trial. Together, these findings imply that psychedelic‑use may causally influence metaphysical beliefs—shifting them away from ‘hard materialism’. Functional diversity holds the promise of understanding ecosystems in ways unattainable by taxonomic diversity studies. Underlying this promise is the intuition that investigating the diversity of what organisms actually do—i.e. their functional traits—within ecosystems will generate more reliable insights into the ways these ecosystems behave, compared to considering only species diversity. But this promise also rests on several conceptual and methodological—i.e. epistemic—assumptions that cut across various theories and domains of ecology. These assumptions should be clearly addressed, notably for the sake (...) of an effective comparison and integration across domains, and for assessing whether or not to use functional diversity approaches for developing ecological management strategies. The objective of this contribution is to identify and critically analyze the most salient of these assumptions. To this aim, we provide an “epistemic roadmap” that pinpoints these assumptions along a set of historical, conceptual, empirical, theoretical, and normative dimensions. (shrink)

The pervasiveness of unethical actions paired with the rising demand for creativity in organizations has contributed to an increased interest in how ethicality and creativity relate. However, there are mixed findings on whether these two fundamental pillars of the workplace relate positively, negatively, or not at all. To provide an empirical consensus to this debate, we study the directional effects of ethicality on creativity by employing meta-analytic techniques. Specifically, a series of meta-regressions, moderated meta-regressions, and individual subgroup analyses of moderators (...) investigated the nuances of the ethicality-creativity relationship. Using a random-effects model, a quantitative review of 278 effects across 23 articles revealed that ethicality positively related to creativity. We further probed how each domain of ethicality (i.e., ethical decision-making, ethical leadership, ethical culture, ethical thought) related to creativity; the effect of ethicality on creativity remained positive, but was no longer significant. Additionally, to expand our understanding of this relationship, we offer theoretical and empirical accounts of five factors that moderate the positive association between ethicality and creativity: ethicality and creativity domains and measurement, sample culture, study characteristics, and sample characteristics. Disability & Justice: The Capabilities Approach in Practice is an interdisciplinary examination of the practical application of the capabilities approach viewed through the lens of the experience of disability. Careful and critical examination of vital foundational concepts is undertaken prior to contextualizing the experience of disability and how we might begin to promote an inclusive society through an application of the capabilities approach.

Generative artificial intelligence (AI) has the potential to transform many aspects of scholarly publishing. Authors, peer reviewers, and editors might use AI in a variety of ways, and those uses might augment their existing work or might instead be intended to replace it. We are editors of bioethics and humanities journals who have been contemplating the implications of this ongoing transformation. We believe that generative AI may pose a threat to the goals that animate our work but could also be (...) valuable for achieving those goals. In the interests of fostering a wider conversation about how generative AI may be used, we have developed a preliminary set of recommendations for its use in scholarly publishing. We hope that the recommendations and rationales set out here will help the scholarly community navigate toward a deeper understanding of the strengths, limits, and challenges of AI for responsible scholarly work. Duchenne's muscular dystrophy (DMD), which affects 1/3500 live male births, involves a progressive degeneration of skeletal and cardiac muscle, leading to early death. The protein dystrophin is lacking in DMD and present, but defective, in the allelic, less severe, Becker muscular dystrophy and is also missing in the mdx mouse. Experiments on the mdx mouse have suggested two possible therapies for these myopathies. Implantation of normal muscle precursor cells (mpc) into mdx skeletal muscle leads to the conversion of dystrophin‐negative fibres (...) to ‐positive, with consequent improvement in muscle histology. Direct injectidn of dystrophin cDNA into skeletal or cardiac muscle also gives rise to dystrophin‐positive fibres. Although both appear promising, there are a number of questions to be answered and refinements to be made before either technique could be considered possible as treatments for myopathies in man. ABSTRACT:This article examines the place of Machiavelli’sPrincein the history of ethics and the history of leadership philosophy. Close scrutiny indicates that Machiavelli advances an ethical system for leadership that involves uprooting corruption and establishing rule of law. He draws on history and current affairs in order to obtain a realistic understanding of human behavior that forms a basis for a consequentialist ethics. While he claims a good leader might do bad things, this is in situations where necessity constrains a prince (...) to choosing the “least bad” course of action. Furthermore, Machiavelli advocates winning the goodwill of followers through leadership as a source of power. Machiavelli’s leadership ethics has a sophistication not fully enjoyed by his reputation in management scholarship. He would not score as especially “Machiavellian” on the Mach IV. Many of his ideas contain seeds for theories that are now considered important for leadership today. (shrink)

La investigación ecológica a largo plazo (Long Term Ecological Research, LTER) maneja problemas que abarcan décadas o plazos más largos. El programa y su nombre formal comenzaron en Estados Unidos en 1980. Si bien los estudios y observaciones a largo plazo comenzaron tempranamente en 1400 y 1800 en Asia y Europa, respectivamente, el enfoque a largo plazo no se formalizó sino hasta el establecimiento de los programas de investigación ecológica de largo plazo en Estados Unidos. Estos programas han permitido experimentos (...) a nivel de ecosistemas y comparaciones entre sitios que condujeron a una mejor comprensión de la estructura y funcionamiento de la biosfera. El enfoque ecosistémico holístico de esta iniciativa permite también la incorporación de la dimensión humana en la ecología, y recientemente ha dado lugar al nuevo concepto de investigación socio-ecológica de largo plazo (LTSER). Hoy los programas de investigación socio-ecológica a largo plazo existen en por lo menos 32 países (esto es, los miembros de la Red Internacional de Investigación Ecológica a Largo Plazo o ILTER (International Long Term Ecological Research). Sin embargo, la consolidación de la red internacional dentro del paradigma de investigación socio-ecológica a largo plazo todavía requiere: (1) inclusión de algunas regiones remotas del mundo que aún están poco representadas, como el suroeste de América del Sur; (2) modificaciones en el tipo de investigación realizado, tales como la integración de las ciencias sociales y naturales con las humanidades y la ética, y (3) la incorporación de las conclusiones y resultados dentro de los procesos sociales y políticos. En este contexto, la naciente red de investigación socio-ecológica a largo plazo en Chile, que se extiende en el rango latitudinal más largo de bosque templado en el Hemisferio Sur, agrega una nueva región remota a las investigaciones ecológicas de largo plazo que había sido pasada por alto anteriormente. Además, la colaboración con la Universidad de North Texas y otros asociados internacionales ayuda a desarrollar un enfoque interdisciplinario para integrar las ciencias ecológicas y la filosofía ambiental, junto con los conocimientos ecológicos tradicionales, la educación informal y formal, la política, las humanidades, los procesos socio-políticos y la conservación biocultural. How can the power of wholes be resisted without essentializing their parts? Drawing on different archives and methodologies, including aesthetics, history, biology, affect, race, and queer, the interventions in this volume explore different ways of troubling the consistency and stability of wholes, breaking up their closure and making them more dynamic. Doing so without necessarily presupposing or producing parts, an outside, or a teleological development, they indicate the critical potential of partiality without parts.

Four pairs of connectionist simulations are presented in which quasi‐regular mappings are computed using localist and distributed representations. In each simulation, a control parameter termed input gain was modulated over the only level of representation that mapped inputs to outputs. Input gain caused both localist and distributed models to shift between regularity‐based and item‐based modes of processing. Performance on irregular items was selectively impaired in the regularity‐based modes, whereas performance on novel items was selectively impaired in the item‐based modes. Thus, (...) the models exhibited double dissociations without separable processing components. These results are discussed in the context of analogous dissociations found in language domains such as word reading and inflectional morphology. (shrink)

A tendency of auditory cortical neurons to respond at the beginning of major transitions in sounds rather than providing a continuously updated spectral-temporal profile may impede the generation of combination-sensitivity for certain classes of stimuli. Potential consequences of the cortical encoding of voiced stop-consonants on representational principles derived from orderly output constraints are discussed.
